Development and validation of analytical methods

Trichinella spp. - Three analytical methods were developed, validated and accredited and are available as part of the services provided by the EURLP to NRLs for parasites: two ELISA test for the detection of anti-Trichinella antibodies in human and swine sera, respectively, and a Multiplex PCR method for the identification of Trichinella parasites at species level. Moreover, the magnetic stirrer method for pooled sample digestion for the detection of Trichinella in fresh meat (Commission Regulation No 2075/2005, see Section "Documents - EU Regulations"), was also accredited.
Anisakidae worms - EURLP developed, validated and accredited a PCR/RFLP method and a Multiplex PCR for the identification at species level of parasites of the family Anisakidae. With these methods is possible to identify parasites belonging to the following species: A. simplex sensu-stricto (ss), A. pegreffii, A. simplex C, A. simplex s.l., A. simplex/pegreffii hybrids, A. ziphidarium, A. physeteris, A. typica, Anisakis sp. A and Pseudoterranova spp, Hysterothilacium aduncum, P. decipiens (or P. krabbrei, or P. bulbosa). The methods were accredited in July, 2010 and July, 2013.
Echinococcus spp. - EURLP developed, validated and accredited a PCR-sequencing method for the identification at species level of parasites belonging to the Echinococcus granulosus complex. The method is able to identify parasites belonging to the following species: E. granulosus sensu strictu (G1, G1BC, G2, G3 genotypes), E. equinus (G4 genotype), E. ortleppi (G5 genotype), E. intermedius (G6 and G7 genotypes) and E. canadensis (G8 and G10 genotypes). The method was accredited in July, 2010.
Cryptosporidium spp. - EURLP developed, validated and accredited a PCR/RLFP method for the identification at species level of parasites belonging to the genus Cryptosporidium. The method is able to identify parasites belonging to the following species: C. parvum, C. hominis, C. canis, C. felis, C. meleagridis, C. andersoni, C. suis, the Cervine and Horse genotypes. The method was accredited in July, 2011.
Giardia duodenalis - EURLP developed, validated and accredited two methods (PCR/RLFP and PCR) for the identification at assemblage level of parasites belonging to the species Giardia duodenalis. The methods are able to identify parasites belonging to the assemblages A, B, C, D, E, F and G, indistinguishable on morphological basis. The methods were accredited in July, 2012 and July, 2013.
Opisthorchis spp. - EURLP developed, validated and accredited an ELISA method for the detection of anti-Opisthorchis sp. antibodies in human serum, and a PCR method for the detection of Opisthorchis sp. eggs in faeces of human and animal origin. The methods were accredited in July, 2012.
